Description

This literature discussion highlights recent research from the Rosenzweig group that probes the structural and spectroscopic properties of a newly discovered, atypical CuA site located in a PmoD protein in a new class of methane-oxidizing bacteria.

This LO is part of a special VIPEr collection honoring the 2021 ACS National Award recipients in the field of inorganic chemistry. Amy Rosenzweig was the recipient of the Alfred Bader Award in Bioinorganic or Bioorganic Chemistry for pioneering work in bioinorganic chemistry elucidating metalloenzyme catalysis and cellular metal homeostasis at the molecular level.

Learning Goals

Students will be able to

  • read the current literature and appreciate the suite of experimental techniques that lead to structural elucidation of this CuA dicopper center
  • interpret optical spectra to understand the ligation around the copper centers and the pathway for CuA formation
  • apply bioinorganic and inorganic concepts to interpret results, such as Hard-Soft Acid/Base theory, amino acid variants, and conditions for formation and cleavage of disulfide bonds
Implementation Notes

This literature discussion requires close reading of the article in order to understand the experimental results. It should be used for upper level students, after discussion of hard-soft acid-base chemistry, coordination chemistry, and optical spectroscopy. Most of the questions can be answered without referencing the EPR data, if that is beyond the scope of your course.
